Figure 2: The cytotechnologist will scan for CTCs showing a combination of CK and DAPI staining but without CD45 staining, while ruling out any artifacts that may be present. Some of these include non-specific junk showing in each channel, which is a similar finding in positive control cells that may carry over to other samples on very rare occasions. Other pitfalls include fuzzy signals where we are unable to confirm the CTC and non-specific signals where a positive DAPI and CK signal is present without CD45 staining, but the DAPI and CK are not seen in the same locations as seen with a viable CTC.
